While we should have a healthy skepticism when it comes to sightings, this lead does seem a little better than others.
It is rather odd that besides a dog's scent of Matt and Sweat, there really hasn't been much of a trail since their escape.
Usually there's a general gist of where an escapee may be. These guys could be anywhere.

I'm assuming there have been numerous sighting reports since the escape, so since this one has gotten media attention, there must be at least a decent degree of certainly that this is accurate. Ha, I posted my response to this new lead before I even read your reply.

"After a tip about two men walking along railroad tracks led 300 federal, state and local law enforcement officers on the hunt for two escaped killers to this rural Allegany County town over the weekend, a new lead arose that the missing prisoners may still be near the prison. And this time, there’s forensic evidence. A law enforcement source close to the investigation told The Buffalo News that Saturday someone reported seeing two men who matched the descriptions of Richard W. Matt and David P. Sweat near Clinton Correctional Facility in Dannemora, from which they escaped two weeks ago. Then, on Sunday afternoon, “forensic evidence” that Sweat was in the vicinity of the sighting was confirmed." So we have a promising but not confirmed lead about Western NY near the PA border (hey MSNBC, misinformation is CNN's job! (Just kidding CNN, I love that you air The Hunt!). But also this forensic evidence that proves they were more towards the prison two weeks ago. Of course, the old advice still rings true. Update: DNA from at least one of the two escaped New York prisoners was found at a burglarized cabin in a rural town about 20 miles from the prison, sources told NBC News on Monday. The search for the men, Richard Matt and David Sweat, focused on the town of Owls Head after the DNA was found on Saturday. Police announced they were wrapping up their search in southwest New York, near the Pennsylvania state line, where a witness had reported spotted two men on a railroad line. About 300 law enforcement officers searched the area and found it to be clear. This is promising. They may be closing in, little by little, as it appears Joyce Mitchell not showing up as their getaway driver did cause them to have to wing it and not be able to immediately flee the vicinity.

Prison-issued underwear was found at the cabin in Owls Head. So the past few major fugitive hunts have now involved Eric Frein and his diapers, and Richard Matt/David Sweat and their prison underwear lol. But hey, whatever helps to get them captured. www.classichitsandoldies.com/v2/63499/

He was commentating on CNN too. While I wish the circumstances were different and it wasn't because of two vicious killers on the loose putting neighboring upstate towns on edge, it's really cool to see people involved with AMW still commentating on tv. (I love The Hunt but I still hold out an admittedly naive hope we will one day see John Walsh on CNN while another crime fighter takes up his mantle hosting AMW on another network.)

I'm glad for the media staying on top of the story too, although sadly I don't think they would have if it weren't for the role of Joyce Mitchell in the escape. She provided a sensationalist aspect- allegedly helping Matt and Sweat escape, possibly having sexual relations with one or both of them- that the media ate up and ran with. Sadly, if this were a "normal" escape, nothing planned, and the two just happened to jump a fence while no one was watching them- I'm not so sure it would still be front page news. For the first few days maybe, but the media seems more obsessed with Mitchell than the actual murderers still on the loose at times. At the same time, anything that keeps these two in the public eye will helpfully lead to their capture.
